i would recommend 2 holes in case a snail or algae blocks one you have a back up also make sure the hole is pointing down so you not spraying water out of your tank

I got holes in both of my loclines and I make sure to see that they're clean when I do a water change, if not there will be flood.

I have anti siphon holes on my stand pipes, not return though. But then again my sump is plumbed to account for the extra water
